She's 23 and she likes quirky platformers and games influenced by Eastern Asia. I know a lot more about the former and am leaning towards de Blob 2. However, I do not know much about the latter and would be open to suggestions that are potentially better than my idea

Okamiden may be up her alley in terms of what it is about. I don't know about save point frequency and how long cutscenes last. Those could be deal-killers if they aren't at appropriate levels

My first thought was The Legendary Starfy, thinking that this was for a little girl. Even though I was wrong, after seeing her taste in games, I stand by my pick.

Umihara Kawase Shun Second Edition?
It's an import and obviously not in english but it's a cult physics based platformer where you hook onto objects with a fishing pole and swing/bounce off of them to get past obstacles. There's multiple routes through the game, and multiple ways of approaching levels based on your skill level. And there's a replay feature. Also I think it's supposed to have all the levels from the SNES and PSX versions of the game.

I guess there's also Solatorobo: Red the Hunter, although that kind of skews towards a younger demographic and is rather simple/easy.

She actually inquired about Professor Layton. I am only worried that there will be plenty of puzzles that will frustrate her to the point of destroying property and/or quitting (I'm looking at those games where you have to rearrange blocks so that one object moves from one end to the other).
Otherwise it would be the clear-cut #1 choice. Right now I am considering (in no order):
1. de Blob 2 2. Kirby Mass Attack 3. Kirby Superstar Ultra 4. Professor Layton title (probably will be the one coming out soon) 5. Okamiden 6. Legendary Starfy

Could you guys explain The World Ends With You a bit more in detail? I know that I dismissed that as an option at first but that's only because I heard that you fight two separate battles on two screens.
Is that true? If so, how simple is it? My sister won't mind difficult games but complexity is another thing. If it's too complicated, it will just be a waste of time to her

There are two characters and they share the same HP bar. You control the main character with the touch screen. The partner is on the top screen, and they attack/dodge based on pressing the control pad/buttons to attack and dodge. They pass a light baton between them as they successfully land attacks and the one holding it does more damage or something so you can tell which screen to focus on.
Though you can set the partner to auto control, and you can adjust the difficulty level at any time to make things as easy or hard as you like (and there's different item drops on various difficulty levels and you can increase the drop rate by temporarily lowering your level, so there's reasons to do this)
