The more votes, the better. Only rule I ask is: Please do not limit your list to entirely one genre. I'd like a little variety on there.

Reasons for this topic's existence will be kept confidential until a later time. You will see (:

Crouching Tiger, Hidden Dragon
Mulholland Drive
Shrek
Spirited Away
Love, Actually
The Lord of the Rings: Return of the King
Pans Labyrinth
Casino Royale
The Dark Knight
Up

I absolutely hate Love, Actually but I wont deny that it was definitive. Shrek is there because it basically invented the modern non-Pixar animated formula and is the reason the Best Animated Feature oscar exists. The rest are fairly self-explanatory. No Avatar because it came out with days left in the decade and nearly all its impact was felt in the 2010s.

Definite "definitive of the 2000s". The best films, straight up? The films most indicative of 2000s thematic elements and style? The most culturally relevant films of the decade?

Honestly, though:

The Matrix ('99 but is -the- definitive millennium-era film.)
Shrek (the success of which both spurned the decline of 2D animation and a veer towards subversive, cynical storytelling that can still be felt today. Everyone wanted to have a Shrek.)
The Lord of the Rings: The Fellowship of the Ring
Mean Girls
Kill Bill Part I
The Fast and the Furious (huge contributor to action film sequelitis into the 2010s)
Ocean's Eleven
No Country for Old Men
Twilight
The Dark Knight

When I hear your question, what comes to mind first is impact during the decade itself (admittedly an inherently biased metric), with a side of lasting pop-cultural position and perception. As such, my choices based on those criteria are, in alphabetical order:

Crouching Tiger, Hidden Dragon
Finding Nemo
Gladiator
Harry Potter and the Sorcerer's Stone
Pirates of the Caribbean: The Curse of the Black Pearl
Shrek
Spider-Man
Star Wars: Episode III - Revenge of the Sith
The Dark Knight
The Lord of the Rings: The Fellowship of the Ring

I would quite like to shout out my (reasonably well-known) favorites instead, but ah well.

Lord of the Rings 1
Lord of the Rings 2
Lord of the Rings 3
Harry Potter 1
Mulholland Drive
Mean Girls
Brokeback Mountain
No Country for Old Men
The Dark Knight
Ratatouille

LotR is lauded enough by critics and fans, financially successful enough, and culturally impactful enough (then and now) that it deserves 30% of the list - you can't omit any of the trilogy. Harry Potter isn't quite as impactful (at least in cinematic form), but the series was still big enough that it deserves one representative.

Brokeback Mountain was a watershed film - if made today, it would hardly be considered controversial, which illustrates the stark difference in LGBT acceptance as recently as when the movie actually released in 2005.
