If you absolutely want the full picture, you only need to read The Stand, the Low Men in Yellow Coats portion of Hearts of Atlantis, and Jerusalem's Lot. There are references in most of his works, but those are the only three that would make a huge impact. Even then, I'd argue that you only really need Low Men In Yellow Coats. The Stand is great and gives the longest look at a Dark Tower character, but you get the idea just from TDT.
