What do you think is the funniest moment in an otherwise serious film? Like, a big laugh that comes out of nowhere. Not counting borderline stuff like MCU and action blockbusters which are basically secondary comedies.

William Hurt fumbling with his keys in A History of Violence. I don't know if I'd call it the funniest, but for some reason it was the first thing that popped into my head as a moment of uncharacteristic levity in an otherwise dead-serious movie.

In an older WW2 movie, a bunch of soldiers are huddled in trench when an enemy plane flies over themand drops propaganda leaflets instead of bombs. One of the soldiers collects a bunch of them and says with an impish grin on his face that he's very glad to for the delivery of much-needed TP and goes off to do his natural duty.
He dies in an artillery strike a scant few minutes later...
