I think you could do a pretty good genre blend

like just have a bunch of independent stages and they all play differently

One of the very earliest post-Dragon Quest JRPGs was Glory of Hercules, which was loosely based around his labours and involved Aphrodites being trapped in the underworld. I never actually got through it, though, as its very primitive. The second one was much more playable, and the third is one of the better RPGs on the SFC. I have yet to play the 4th one, though it sounds great, and the DS game was boring.

Probably, though I don't know what kind of gameplay would be needed for the Labours to smoothly translate over.

For one thing, I have no idea how you would properly translate the eleventh Labour, the Golden Apples of Hesperides, into a video game and make it be interesting. How is Hercules holding the Heavens in Atlas' stead going to be handled? The only thing I could think of would be a series of QTEs/button inputs with more than a few failures being instant Game Overs, and that can't be fun in any possible metric.

Same goes for the sixth Labour(or was it fifth?), the Augean Stables. How are you going to depict changing a river's course in a video game? Sudden genre-change into something Sim City-esque? Just have him move and break blocks or whatever?

At least the rest of the Labours have a major 'adversary', or at least a bunch of conflict(e.g. the ninth Labour, where Hercules massacres a bunch of Amazons at the end) to focus on for a game.
