Started with Commando since he was the first option. Should I beat the game with him and max out the skill trees?

You would need to beat the game, like three times if you want to come close to maxing out all three skill trees.

Just make a new character whenever. You can switch back and forth through characters you feel like playing as with ease from the menu.

Yeah you have to do three play throughs to reach the max level of 72.

You can always go back and switch to see who you like best. Personally I liked the siren and psycho the most.
