It would be pretty hard. Biggest problem is getting Columbo assigned to the case. He's a homicide detective so at first he wouldn't even be investigating since Light made it all look like heart attacks. He's solved some "impossible" murders before though like the one where the murderer used a robot to pretend to play a wargame to set his alibi and he is religious so he probably wouldn't immediately discount a supernatural murder if there was evidence of it.

If he ever somehow managed to track down Light in person though it's over. He's super perceptive and would immediately be able to pick up on Light's social cues that the book was really important and he has a habit of grabbing shit that is relevant to murder even if he has no idea what it is (see: Suitable to be Framed). Because nobody knows his first name and he refuses to give it when asked, Light could not use the Death Note on him.
