Yes, higher difficulty and more skulls increase your point multiplier. Of course you actually have to beat the level with the drawbacks tho, and Reach is one of the Harder Halo games

Heroic with as many skulls as you can handle or Legendary with only a couple or so skulls

Basically pick a difficulty you are comfortable with, and whatever skulls you think you can survive on + Iron Skull.

Iron Skull increases multiplier more than the other skulls by a lot iirc
