My favorite has to be TeamFourStar's Pokemon Leaf Green Nuzlocke. I don't think I've ever laughed harder while watching a Let's Play than when the gang fought (and then caught) Bork Laser the Snorlax. Their reactions were fucking priceless. *volume warning at the beginning of the video below*

I loved the "Uno: The Movie" Let's Play Achievement Hunter did on the Let's Play channel with modified rules that had it going for almost 3 hours and everyone doing it ended up hating it.
At least up until the end where they pretty much just threw the game so it would be over and could go home. Didn't really care for that because it kinda ruined the point of it. I marked the spoilers if you'd actually care about such things in a Let's Play.

A lot of Vinesauce stuff. The moments that occur to me at the moment are:

- LotR-based text game. Joel(as Sam) gets Frodo high on pipe-weed, then bashes his brains out because he's 'hogging the bong'.

- Link: The Faces of Evil. Joel spends like 40 minutes grinding for snowballs and fireballs and dying a zillion times to scale a waterfall stage. His reward at the end: A life upgrade. He cries in joy... then jumps off a cliff to commit suicide by mistake(and screams; mood whiplash lol) and has to start all over again.

- Shadow President. Joel nukes Norway for no reason, calls his cabinet pussies when they all resign in protest, then gets shot in his office.

Giant Bomb's playthrough of Phantasmagoria. There's one particular scene where the demon possessed husband, 'rapes' the protagonist. But it's so poorly acted and ridiculous, it ends up being hilarious. Vinny and Dave's reactions make it even better.

Also their playthrough of Contradiction is pretty good. You can see some of it in this best of video.

TFS is great, but one that sticks out to me is Minotaur Zombie and his buddy playing Secret of Evermore.

Early on in the game, you're told to go to the hut of Strongheart, a guy you rescued. He gives you the Cure (Remove status ailments) alchemy. They skipped this and ended up finding a secret that nobody else on the forums were aware of - the fish vendor in the second world, an innocuous NPC nobody cares about, gives it to you.

Vinesauce Joel does have a lot of great moments. I like that his stuff is live too. The Lion King bootleg was pretty hilarious.
