All 2nd run does is skip the intro and change a couple of minor details. No plot change. Also if you care about ranks, RE2make is surprisingly generous. S+ Rank on Hardcore is just: Save Less Than 3 times, Beat the game in under 2.5 hours and don't use infinite cheat weapons.
