It'd be vastly more likely to disable it via using magic to redirect some of the force (maybe to throw him away from the blast radius), to rip open a portal to hide behind, to not even be in the area in the first place, via misdirection. In current books, it takes a really powerful hit to sucker punch Dresden out.

There are ways Megumin could win with her one shot, but the odds are way stacked against, due to the nature of the fight.
