Even in the fucking manga it was explained that Yamato using male pronouns was because 1.) to honor the man who stood up to her abusive father and 2.) she was a victim of abuse herself by said father because he wanted a son. The argument for Yamato being a trans man is that he refers to himself as a man, uses male pronouns, everybody around him uses male pronouns and acknowledges him as "Kaido's son", and he prefers to join the other men in gendered activities such as which public bath to use.
