If we are strictly going by the definition of "best bro" then Junpei is objectively the worst. He spends half the game being a jealous little bitch towards the MC

It's a toss-up between Ryuji and Yosuke for best, but I would give it to Ryuji. He gives off the biggest "ride or die" vibes and seems like he would go to war with you any day. Yosuke seems more like he would try to weasel out of it, and maybe not defend you quite as hard

Edit: Another point towards Ryuji is that he is very much "bros before hoes", like when he BTFOs Ann in her social link. Yosuke is girl crazy and Junpei cares more about Chidori
