What is a guillotine league?
The differences with guillotine league are:

• No head-to-head match-ups
• The standings are ranked each week by score. The lowest score each week is eliminated permanently.
• 18 teams

What are the league settings?

• 14-man roster
• Start 1 QB, 2 WR, 2 RB, 1 TE, 2 Flex
• 1.0 PPR
• 6 teams make playoffs, playoffs start week 15

My first time using Sleeper as well, but seems pretty straightforward. Sleeper doesn't have a guillotine setting, so I'll be manually dropping the lowest team each week.
