I think it'd be a neat concept to set it a few decades after the end of ME3. Base the plot around discovering what happened to Shepard in the aftermath of the Reapers' destruction, let us see a new galactic society forming out of the ashes of the Reaper war. You could even have the old squad appear. A grizzled, older Garrus, Tali as a Quarian admiral, an older Jack still teaching young biotics, that sort of thing.
