I'm getting this topic up rather early since I will want to get in some sleep before it begins, yet I also want it up early so people are aware of it.
The Nintendo Direct will be around 40 minutes long and feature information on numerous upcoming games such as Animal Crossing. There may even be more information for Pokemon Sword/Shield, Super Mario Maker 2, and information on the next fighter DLC character for Smash Ultimate. Metroid Prime 4 is not expected to be shown or even mentioned as the game is still in the early development stages.
Treehouse Live starts immediately after the press conference has finished. Day 2 and Day 3 of Treehouse Live will most likely start at the same time (12:00 PM EST) on their respective days, though I can't find any information regarding the starting time for Day 2 and Day 3. Although technically a Nintendo Direct, I called it a press conference in the topic title since saying "Nintendo's Nintendo Direct" seems really awkward. This topic may also be used to discuss Treehouse Live.

Nintendo's Twitch account will also stream Day 2 and Day 3 of Treehouse Live, but you can use the e3.nintendo.com link to ensure that you are able to watch all 3 days of Treehouse Live coverage. The e3.nintendo.com link uses the Youtube live streams. Nintendo has not yet scheduled their Day 2 and Day 3 Treehouse Live streams on Youtube yet, so I can't provide links to them in this post.

I think we'll get two Smash characters, especially since they want all of the DLC characters to be released by early next year.

One character to release now or this week, and announcing the next for release in a month or two. Seems entirely plausible. I wouldn't even be surprised if they announce all four together.
